Understanding the heart's rhythm

June 11, 2013 ? The heart's regular rhythm is crucial to the delivery of oxygenated blood and nutrients to all the organs of the body. It is regulated by a bundle of cells called "the pacemaker," which use electrical signals to set the pace of the heart. Dysfunction in this mechanism can lead to an irregular heartbeat, known as arrhythmia, and often necessitates the implantation of an artificial pacemaker.

Previously, scientists found that many cases of inherited arrhythmias originating in the pacemaker could be attributed to functional defects in the channels responsible for the flow of sodium and calcium. Now Prof. Bernard Attali of Tel Aviv University's Sackler Faculty of Medicine and his fellow researchers have discovered a previously unidentified potassium channel in the cardiac pacemaker which helps to regulate the heartbeat. He hypothesizes that some cases of unexplained arrhythmia could be traced back to irregularities in this channel.

Developing therapies to target this potassium channel could be a significant step towards circumventing artificial pacemakers in favor of biological options, says Prof. Attali. This research has been reported in the journal PNAS.

A cellular heart model

To further investigate the workings of the biological pacemaker, Prof. Attali and his fellow researchers turned to embryonic stem cells isolated from human subjects. Once coaxed into differentiating into cardiac tissue, these cells began to beat automatically, like a small human heart.

While observing and recording the cells' electrical activity, researchers discovered the existence of a new channel in the pacemaker. Facilitating the flow of potassium from the pacemaker cells, this channel triggers the repolarization of the cells -- returning the cell membrane from a "beating" to a "resting" state -- and automatically renews or "restarts" the cycling of the heart.

Since discovering this channel in the embryonic heart, the researchers have shown that the channel exists in the adult heart as well. This finding deepens medicine's understanding of the heart's pacemaker function, which has been the subject of scientific research for over a century.

Screening for mutations

The next step is to conduct screening for mutations in the gene encoding the potassium channel, a process already underway at the TAU-affiliated Sheba Medical Center. "We would like to understand if there are genetic diseases linked to this channel," such as a previously unknown cause of arrhythmia, explains Prof. Attali. If a mutation is found, researchers can begin the hunt for drug compounds, which target this channel. The ultimate goal, he adds, is to be able to treat heart arrhythmias biologically by altering the properties of the pacemaker bundle, rather than relying on a human-made electric pacemaker.

One possible solution could be transplanting healthy pacemaker cells -- developed from a patient's own stem cells -- to replace dysfunctional cells and restore proper heart rhythm. This method would circumvent a common risk of the body rejecting a mechanical transplant.

Video gamers really do see more: Gamers capture more information faster for visual decision-making

June 11, 2013 ? Hours spent at the video gaming console not only train a player's hands to work the buttons on the controller, they probably also train the brain to make better and faster use of visual input, according to Duke University researchers.

"Gamers see the world differently," said Greg Appelbaum, an assistant professor of psychiatry in the Duke School of Medicine. "They are able to extract more information from a visual scene."

It can be difficult to find non-gamers among college students these days, but from among a pool of subjects participating in a much larger study in Stephen Mitroff's Visual Cognition Lab at Duke, the researchers found 125 participants who were either non-gamers or very intensive gamers.

Each participant was run though a visual sensory memory task that flashed a circular arrangement of eight letters for just one-tenth of a second. After a delay ranging from 13 milliseconds to 2.5 seconds, an arrow appeared, pointing to one spot on the circle where a letter had been. Participants were asked to identify which letter had been in that spot.

At every time interval, intensive players of action video games outperformed non-gamers in recalling the letter.

Earlier research by others has found that gamers are quicker at responding to visual stimuli and can track more items than non-gamers. When playing a game, especially one of the "first-person shooters," a gamer makes "probabilistic inferences" about what he's seeing -- good guy or bad guy, moving left or moving right -- as rapidly as he can.

Appelbaum said that with time and experience, the gamer apparently gets better at doing this. "They need less information to arrive at a probabilistic conclusion, and they do it faster."

Both groups experienced a rapid decay in memory of what the letters had been, but the gamers outperformed the non-gamers at every time interval.

The visual system sifts information out from what the eyes are seeing, and data that isn't used decays quite rapidly, Appelbaum said. Gamers discard the unused stuff just about as fast as everyone else, but they appear to be starting with more information to begin with.

The researchers examined three possible reasons for the gamers' apparently superior ability to make probabilistic inferences. Either they see better, they retain visual memory longer or they've improved their decision-making.

Looking at these results, Applebaum said, it appears that prolonged memory retention isn't the reason. But the other two factors might both be in play -- it is possible that the gamers see more immediately, and they are better able make better correct decisions from the information they have available.

To get at this question, the researchers will need more data from brainwaves and MRI imagery to see where the brains of gamers have been trained to perform differently on visual tasks.

Old Mars rover makes rock discovery

Nasa's ageing Opportunity rover on Mars has just made what may be one of its most significant discoveries to date.

The nine-year-old robot has identified rock laden with what scientists believe to be clay minerals.

Their presence is an indication that the rock, dubbed Esperance, has been altered at some point in the past through prolonged contact with water.

Opportunity has seen a clay-bearing outcrop before but scientists say this is by far the best example to date.

"It's very rich," said Steve Squyres, the rover's principal investigator.

"We've been discovering evidence for water on Mars since we first landed back in 2004. What's different here?

"If you look at all of the water-related discoveries that have been made by Opportunity, the vast majority of them point to water that was a very low pH - it was acid.

"We run around talking about water on Mars. In fact, what Opportunity has mostly discovered, or found evidence for, was sulphuric acid.

"Clay minerals only tend to form at a more neutral pH. This is water you could drink. This is water that was much more favourable for things like pre-biotic chemistry - the kind of chemistry that could lead to the origin of life."

Prof Squyres, who is affiliated to Cornell University, Ithaca, New York, said he was inclined to put Esperance in his personal top five discoveries made on the Red Planet by Opportunity and her twin rover, Spirit, which stopped working in 2011.

The clays are aluminium-rich, possibly of the type montmorillonite. However, because Opportunity's X-ray spectrometer can only discern the atomic elements in a rock, and not their mineralogical arrangement, no-one can say for sure.

Nonetheless, the mere occurrence of clays is further proof that Mars was much warmer and wetter billions of years ago; a very different place to the cold, desiccated world it has become.

And these results complement nicely those of Nasa's newer rover Curiosity, which has also identified clays at its landing site almost half-way around the planet's equator.

The old robot made its find at a location called Cape York, which is sited on the rim of a 22km-wide crater known as Endurance.

Mission managers have now commanded it to start moving along the ridge to a destination dubbed Solander Point.

There is an expectation that Opportunity will find a deeper stack of rocks at the new location to follow up the Esperance water story.

"Maybe [we can] try to reconstruct the actual depositional environment of these materials and whether they were lacustrine - that is, formed by a lake - or fluvial (river) or an alluvial fan (network of streams), or whatever," said deputy principal investigator Ray Arvidson, of Washington University, St Louis.

'Daily gift'

Opportunity is now operating well beyond its expected lifetime.

When it landed at Eagle Crater in January 2004, Nasa hoped to get at least 90 working Martian days (sols) from the machine. Remarkably, it continues to roll beyond 3,300 sols.

It has an "arthritic" robotic arm, its solar panels are losing efficiency, and it drives backwards to save wear on its locomotion system.

It is also now having to contend with glitchy flash memory. But the US space agency is determined to keep pushing the vehicle for as long as possible.

"Remember, the rover continues in a very hostile environment on Mars," said John Callas, Nasa's Opportunity project manager.

"The rover could have a catastrophic failure at any moment. So, each day is a gift."
